چکیده

Abstract Background The therapeutic effect of mesenchymal stem cells (MSCs) from human adipose tissue on renal interstitial fibrosis has been demonstrated by several groups. However, the way to enhance renoprotective adipose-derived (AMSCs) and possible mechanisms are still unclear. present study aimed determine whether glial cell line-derived neurotrophic factor (GDNF)-modified AMSCs hold an enhanced protective fibrosis. Methods were isolated purified for culture. gene GDNF constructed transfect into AMSCs. ability GFP-AMSCs GDNF-AMSCs supernatants promote tube formation endothelial cells, repair damaged junctions, improve function was compared using assay, immunofluorescence techniques, vascular ring respectively. Furthermore, HE Masson staining used observe histological morphology kidney in vivo. Peritubular capillary changes detected analyzed fluorescence microangiography (FMA). Meanwhile, hypoxia, oxidative stress, fibrotic markers, PI3K/Akt pathway proteins measured western blot or qRT-PCR technics. Results Compared with only, could injured angiogenesis through secreting more growth factors supernatant GDNF-AMSC culture media vitro studies. Studies vivo, unilateral ureteral obstruction (UUO)-induced mice injected transfected their tail veins. We showed that homing observed group GFP-AMSC group. animals treated exhibited improvement rarefaction induced obstructed we reported protect tissues against microvascular injuries via activation signaling pathway. Therefore, further ameliorated suppressed finally inhibited transition noting decreased coexpression (CD31) myofibroblast (a-SMA) markers. Conclusion Collectively, our data indicated enhances improving microcirculation PI3K/Akt/eNOS afterward inhibit EndMT process fibrogenesis, which should have a vast implications designing future remedies chronic disease (CKD) treatment.

Enhanced renoprotective effect of HIF-1α modified human adipose-derived stem cells on cisplatin-induced acute kidney injury in vivo

Current therapeutic options for acute kidney injury (AKI) are limited to the use of supportive measures and dialysis. A recent approach that has sparked great interest and gained enormous popularity is the implantation of stem cells to repair acutely damaged kidney organ. Hypoxia inducible factor-1α (HIF-1α) is effective in protecting the kidney from ischemia and nephrotoxicity. In this study, ...

Enhanced renoprotective effect of IGF-1 modified human umbilical cord-derived mesenchymal stem cells on gentamicin-induced acute kidney injury

The therapeutic action of umbilical cord-derived mesenchymal stem cells (UC-MSCs) against acute kidney injury (AKI) has been demonstrated by several groups. However, how to further enhance the renoprotective effect of UC-MSCs and improve the therapy effect, are still unclear. In this study, we mainly investigated whether insulin-like growth factor-1 (IGF-1)-modified UC-MSCs hold an enhanced pro...

Regenerative Effect of Bone Marrow-derived Mesenchymal Stem Cells in Thioacetamide-induced Liver Fibrosis of Rats

The present study determined the regenerative effect of bone marrow-derived stem cells (BMSCs) on thioacetamide (TA)-induced liver fibrosis in rats. A total of 30 male Wistar rats were randomly divided into sham control and treatment groups. The rats of the sham control group were subdivided into three groups and sampled on the 14th, 18th, and 20th weeks after fibrosis induction. The rats of th...
